Transformative Proteomics: Leveraging Affinity Based Assays to Advance Immunotherapy and Precision Prevention in Cancer Research

Affinity-based proteomics advances are revolutionizing cancer immunotherapy by identifying disease biomarkers and understanding the pathways that drive the disease. These insights inform personalized therapies and uncover new strategies to improve disease monitoring and treatment resistance. Studies have utilized various proteomics methods to investigate the molecular mechanisms underlying cancer growth, metastasis, and drug resistance, providing valuable insights for personalized therapy. Moreover, proteomics approaches have identified characteristics of drug-resistant cancer cells, suggesting new therapeutic targets and strategies to overcome resistance. In this session we will delve into the dual role that inflammation plays in cancer, promoting antitumor immunity while also driving immunosuppression and resistance to anti-PD-1 therapy in NSCLC. We will also highlight the associations of 5,420 pre-diagnostic plasma proteins measured using the Olink HT panel with prostate cancer risk, using data from the European Prospective Investigation into Cancer and Nutrition Cohort [EPIC]. A fixed-effect meta-analyses was also performed to combine associations estimated in our study with our two previous prospective investigations (one in UK Biobank with data on 2,822 proteins [Olink Explore 1 and 2 panels] in 1,154 cases and one in EPIC with data on 359 inflammation-related proteins [Olink Explore Inflammation 1. Our study uncovered several novel protein associations with prostate cancer risk and provided links for some to metastasising tumour clones.
