NPX Signature
Quality control is done using Olink NPX Signature software. It ensures reliable data by using three internal controls in all samples and external controls in each run.
Line-in-chip detection checks for assays with missing signal in the qPCR. If there are more than five in a sample, the sample is considered failed.
Sample Evaluation: Ensures that internal controls are measured consistently across all samples. Incubation Control and Detection Control should not deviate more than ±0.3 NPX from the plate median. Samples outside of specification get a warning flag in Sample QC.
Assay Evaluation: Based on external controls. All products require:
- minimum two valid Negative Controls
- minimum two valid Calibrators / Interplate Controls (IPC)
- CV less than 30% for Calibrators / IPC
In addition, Target 48, Flex and Focus require minimum two valid Sample Controls
- CV less than 30% for the Sample Controls
- Sample Control accuracy median less than 30% from expected concentration values
Assay is failed if the minimum number of valid Calibrators / IPC replicates is not reached. For all other assay QC requirements, the assay gets a warning flag in Assay QC.
Run Evaluation: All products require:
- minimum two valid Negative Controls
- minimum two valid Calibrators / IPC
- number of flagged or failed assays are less than 1/9 of assays
- number of flagged or failed samples are less than 1/6 of customer samples
- variation in internal controls must have median absolute deviation less than
- 0.23 for customer samples
- 0.25 for external controls
In addition, Target 48, Flex and Focus require:
- minimum two valid Sample Controls
All run QC requirements generate a warning flag in the Run QC. RunQC flags are only shown in UI and not in the exported data.
